Basiliximab is a chimeric (murine/human) monoclonal antibody (IgG1k) which functions as an immunosuppressive agent via specifically binding to and blocking the interleukin-2 receptor alpha-chain (IL-2Rα), also known as CD25 antigen, on the surface of activated T-lymphocytes. The specific, high affinity binding of basiliximab to IL-2Rα competitively inhibits IL-2-mediated activation of lymphocytes, thereby downregulating cellular immune pathways involved in allograft rejection.
As with the similarly targeted, humanized monoclonal antibody daclizumab, basiliximab is indicated
for the prophylaxis of acute organ rejection in renal transplant
patients. Adverse effects include post-administration anaphylaxis, for which a US FDA warning advisory was issued in 2000.
